ABSTRACT

ObjectiveTo explore the quality differences between steamed products and raw products of Citri Reticulatae Pericarpium(CRP). MethodThe color of steamed products and raw products of CRP was determined from the perspective of appearance by electronic eye technique, and the quality differences between them was objectively characterized by the luminous value(L*), yellow-blue value(b*), red-green value(a*) and total chromatic value(E*ab). Based on this, ultra-high performance liquid chromatography(UPLC) was used to establish a fingerprint evaluation method with the mobile phase of acetonitrile(A)-0.1% formic acid aqueous solution(B) for gradient elution(0-5 min, 5%A; 5-30 min, 5%-20%A; 30-60 min, 20%-52%A), detection wavelength at 270 nm, flow rate of 0.3 mL·min-1 and column temperature of 30 ℃. The quality differences between steamed products and raw products of CRP were compared from the perspective of chemical composition, and correlation analysis was used to reveal the correlation between the difference in appearance color and the difference in internal chemical composition. ResultAfter being steamed, L*, b* and E*ab of CRP showed an overall decreasing trend, indicating that the color of the steamed products darkened and deepened from yellow to blue but still tended to be yellow, while a* showed an overall increasing trend, indicating that the color of the steamed products tended to red. A total of 24 peaks were identified in the fingerprint profiles of raw products and steamed products of CRP, and 13 of the main peaks were identified. The precision, stability and repeatability studies showed that compared with the reference peak (peak 14, hesperidin), the relative standard deviations(RSDs) of the relative peak area and relative retention time of the remaining peaks were<3.0%.The results of chemometric statistical analysis showed that there were some differences between raw products and steamed products of CRP, and 7 main differential components were identified, among which 5-hydroxymaltol(peak 1) and 5-hydroxymethylfurfural(peak 2) were the characteristic components of steamed products. The correlation analysis results showed that, in addition to the above two characteristic components, four components of peak 4, peak 10 (vicenin-2), peak 23 (tangeretin) and peak 24 (5-demethylnobiletin) also correlated significantly with the color change (E*ab) of the samples (P<0.05, P<0.01). ConclusionBefore and after steaming, not only the chemical composition changes, but also the color. Comparing the characteristic peaks of chemical composition difference and color difference before and after steaming of CRP, it is found that 5-hydroxymaltol, 5-hydroxymethylfurfural and peak 4 are common characteristic difference components, which can provide a reference for establishing the characteristic quality control method of steamed products, and quickly evaluating the quality difference between raw products and steamed products of CRP.

ABSTRACT

Objective:To develop a novel, flexible, dual-arm, master-slave digestive endoscopic minimally invasive surgical robot system named dual-arm robotic endoscopic assistant for minimally invasive surgery (DREAMS) and to evaluate its feasibility for endoscopic submucosal dissection (ESD) by using ex vivo porcine stomachs.Methods:A novel endoscopic robot (DREAMS) system was developed which was composed of a flexible two-channel endoscope, two flexible robotic manipulators, a master controller, a robotic arm, and a control system. A total of 10 artificial round-like lesions with diameters ranging from 15 to 25 mm were created (5 in gastric antrum and 5 in gastric body) by using fresh peeled stomach of healthy pigs as the model. Submucosal dissection was performed with the assistance of the DREAMS system by two operators. The main outcome was submucosal dissection speed, and the secondary outcomes included muscular injury rate, perforation rate, and grasping efficiency of the robot.Results:All 10 lesions were successfully dissected en bloc by using the DREAMS system. The diameter of the artificial lesions was 22.34±2.39 mm, dissection time was 15.00±8.90 min, submucosal dissection speed was 141.79±79.12 mm 2/min, and the number of tractions required by each ESD was 4.2 times. Muscular injury occurred in 4/10 cases of ESD. No perforation occurred. Conclusion:The initial animal experiment shows the DREAMS system is safe and effective.

ABSTRACT

Objective:By calculating the efficiency of the scientific research laboratory, which reflects the level of the scientific research input and output capacity, provide reference for the evaluation and decision-making of its scientific research sustainable development capacity.Methods:20 scientific research laboratories in a hospital were selected as the research subjects, annual input data were used as the input index. Weighted quantitative scores of the performance of each laboratory in research capacity and contribution, research team construction, discipline development and personnel training, operation management, papers and monographs, patents and transfer, awards, graduate-student training, standards and norms, and academic conferences. All these factors mentioned above were used as output indicators. Then Analytic Hierarchy Process (AHP) and Data Envelopment Analysis (DEA) are used to evaluate the scientific efficiency of each laboratory.Results:The performance of the laboratory is weak in the aspects of patent transfer, awards, standardization. The technical efficiency of laboratory 20 is the lowest, and the scale efficiency of laboratory 12 is the lowest.Conclusions:Scientific Research Laboratories should enhance the effectiveness through input adjustment and output enhancement, meanwhile each laboratory should pay attention to the transformation of scientific achievements and also the optimization of construction system.

ABSTRACT

Objective:To evaluate the diagnostic efficiency of hypersensitivity quantitative fecal immunochemical test (hs-qFIT) in colorectal cancer (CRC) and advanced adenoma.Methods:From July to December 2020, consecutive patients aged 50 to 75 years who underwent colonoscopy in Qilu Hospital of Shandong University, and had the Asia-Pacific colorectal screening score of medium or high risk were enrolled. All patients were requested to complete two hs-qFIT before colonoscopy. The diagnostic efficacy of hs-qFIT for CRC and advanced adenoma were assessed. Receiver operating characteristic curve of hs-qFIT in CRC diagnosis was drawn and the area under the curve (AUC) was calculated.Results:A total of 811 patients including 20 (2.5%) cases of CRC, 47 (5.8%) cases of advanced adenoma, 206 (25.4%) cases of non-advanced adenoma, 219 (27.0%) cases of non-adenomatous polyp, 76 (9.4%) cases of other colorectal lesions and 243 (30.0%) cases of non-colorectal lesions were involved. When the fecal hemoglobin cut-off values were 10, 30, 50, 75 and 100 ng/mL, the positive rates of hs-qFIT detection were 17.9% (145/811), 10.9% (88/811), 8.3% (67/811), 7.4% (60/811) and 5.8% (47/811), respectively. When the cut-off value of fecal hemoglobin decreased from 100 ng/mL to 10 ng/mL, the sensitivity of hs-qFIT for CRC diagnosis increased from 90.0% to 100.0%, and the specificity decreased from 96.3% to 84.2%; and the sensitivity of hs-qFIT for the diagnosis of advanced adenoma increased from 19.1% to 66.0%, and the specificity decreased from 95.0% to 85.1%. The AUC of hs-qFIT for the diagnosis of CRC and advanced adenoma were 0.981 (95% confidence interval ( CI) 0.970 to 0.992) and 0.846 (95% CI 0.807 to 0.886), respectively. When the optimal cut-off values were taken, the sensitivity and specificity were 100.0% and 91.2% for the diagnosis of CRC, and 66.0% and 85.3% for the diagnosis of advanced adenoma, respectively. Conclusion:Hs-qFIT can help the early screening of CRC and advanced adenoma.

Objective:To investigate the expressions of tissue factor (TF) and vascular endothelial growth factor (VEGF) in diffuse large B-cell lymphoma (DLBCL) and their clinical significances.Methods:The clinical data of 80 cases of DLBCL diagnosed at the Second People's Hospital of Lianyungang from January 2010 to December 2017 were collected, and 30 cases of reactive hyperplasia of lymph node (RLN) were selected as the controls. The expressions of TF and VEGF in the two groups were detected by using immunohistochemical staining.Results:The positive rate of TF and VEGF in the DLBCL group was higher than that in the RLN group [TF: 86.3% (69/80) vs. 50.0% (15/30) ; VEGF: 90.0% (72/80) vs. 53.3% (16/30) ; both P < 0.01]. And there was a positive correlation between the expression of TF and VEGF ( r = 0.704, P < 0.05). There was no significant difference in the positive rates of TF and VEGF among the patients with different gender, age and Hans subtypes in DLBCL group (all P > 0.05). The positive rate of TF in DLBCL patients with B symptom, increased LDH, physical status grade ≥2, and extranodal lesion number >1 was higher (all P < 0.05). The positive rate of VEGF in patients with Ann Arbor stage Ⅲ-Ⅳ, B symptom, increased LDH, and extranodal lesion number >1 was higher (all P < 0.05). The positive rate of TF in international prognostic index (IPI) high-risk group was higher than that in low-risk group ( P < 0.01); the positive rate of VEGF in IPI high-risk group and middle-high-risk group was higher than that in low-risk group (all P < 0.01). The expressions of TF ( r = 0.491, P < 0.01) and VEGF ( r = 0.529, P < 0.01) were positively correlated with IPI. The overall survival rates of TF and VEGF low-expression group were higher than those of TF and VEGF high-expression group (both P < 0.05). Conclusion:The expressions of TF and VEGF are highly expressed in DLBCL, which is associated with the IPI. It can provide a reference value in evaluating prognosis of DLBCL.

ABSTRACT

Objective:To understand the statistics of SCI papers in hospitals of Shandong Province during 2009 to 2018.Methods:Web of Science was used to analyze the SCI papar information including the Annual publication volume change, source publications, research directions, collaboration institutions, counries and regions, funding institutions, conferences, as well as highly cited papers.Results:In the past ten years, 27 559 articles were published in total and kept increasling year by year, especially in 2011 to 2015 the uplift was about 30%-40%. All these papers were published in more than 2 700 journals. 23% papers were published in top 10 journals, whose Impact Factors were between 1-5. Papers in the field of oncology counted the most for 23.252%. Shandong University ranked NO.1 regarding collaboration instituions for it has many academically well performed affiliated hospitals as well asteaching hospitals. Doctors in Shandong Province cooperated a lot with the United States and other countries. 56.3% papers were financially assisted by various fundings and the National Natural Science Foundation of China predominated the most. 1 331 papers came from international conferences.There were 75 highly cited papers in the past 10 years.Conclusions:The quantity and quality of SCI papers were increased continuingly because of the guidance and promotion of scientific research policies, as well as the hospitals’ investments. It is necessary to emphasize on the high quality of scholar outputs and improve the scientific research management in the future.

ABSTRACT

Objective:To develop intelligent quality-control system (IQCS) based on deep convolutional neural networks (DCNN), and to prospectively evaluate the clinical feasibility of this system.Methods:Aimed at quality control objectives during gastroscopy such as the observation integrity of gastric mucosal, gastric mucosa visibility, time spent on gastroendoscopy and suspicious gastric cancer detection, four DCNN models including gastroscopic scanning location recognition model, gastric mucosa visibility recognition model, in vivo and in vitro identification model and gastric cancer detection model were designed. A total of 98 385 white light gastroscopy images were retrospectively collected from multiple centers for training and testing the DCNN models. The accuracy, sensitivity and specificity of each model were calculated and the receiver operating characteristic (ROC) curve was drawn. The models were integrated and formed the multi-function integrated IQCS. At the center of gastroendoscopy, Qilu Hospital of Shandong University, 100 consecutive patients who underwent routine gastroscopy were prospectively enrolled. The feasibility of IQCS in real clinical practice was evaluated. The condition of each quality control function of the system (average error point out or correct rate) and the detection of lesions after the examination were recorded. Results:The accuracy, sensitivity and specificity of the model of gastroscopic scanning location recognition to identify each site were 98.40% to 99.85%, 61.95% to 100.00% and 98.65% to 100.00%, respectively; the area under curve (AUC) of ROC curve ranged from 0.997 6 to 1.000 0. The accuracy, sensitivity and specificity of the model of gastric mucosa visibility recognition to identity the mucosal visibility were 97.02% to 98.27%, 85.14% to 99.28% and 93.72% to 100.00%, respectively. The accuracy, sensitivity and specificity of the model of in vivo and in vitro identification were 97.27%, 99.85% and 94.50%, respectively; the AUC of ROC was 0.961 5. The accuracy, sensitivity and specificity of the model of gastric cancer detection were 95.92%, 95.64% and 96.05%, respectively; the AUC of ROC was 0.975 9. The results of feasibility evaluation of IQCS indicated that in the quality control of gastric mucosa observation integrity, the system average error was 0.32 time/case; in the quality control of mucosal visibility, the system average error was 0.47 time/case; the correct rate of intelligent timing during gastroscopy was 96.00%, in the quality control of suspicious gastric cancer detection, the system average error was 0.36 time/case. A total of 3 cases of gastric cancer and 1 case of high grade gastric intraepithelial neoplasia were detected. The system could accurately identify the location. Conclusions:Gastroscopy IQCS can accurately achieve quality control in the observation integrity of gastric mucosa, gastric mucosa visibility, time spent on gastroendoscopy and suspicious gastric cancer detection in actual examination, which makes accurate and efficient gastroscopy quality control possible.

ABSTRACT

Objective To analyze some related causes of hypocytosis or hematocytosis with bone marrow dry pumping. Methods Bone marrow histology, reticular fiber staining and selective immunohistochemical staining were performed in 34 bone marrow dry pumping patients with hypocytosis or hematocytosis in the Second People's Hospital of Lianyungang from January 2012 to August 2017. Results All the patients showed dizziness, fatigue, splenomegaly, night sweats and bleeding, including primary myelofibrosis (17 cases, 50.0 %), chronic myelocytic leukemia (4 cases, 11.8 %), acute myelocytic leukemia (2 cases, 5.9 %), acute lymphoblastic leukemia (1 case, 2.9 %), myelodysplastic syndrome (2 cases, 5.9 %), multiple myeloma (2 cases, 5.9 %), non-Hodgkin lymphoma with bone marrow infiltration (2 cases, 5.9 %), polycythemia vera (1 case, 2.9 %) and bone marrow metastatic tumor (3 cases, 8.8 %). The bone marrow proliferative degree in primary myelofibrosis group was mainly "grade Ⅱ" to "grade Ⅳ", and the non primary myelofibrosis group was mainly "grade Ⅲ" to "grade Ⅴ", and the differences of proliferative degree component between them were statically significant (χ2= 12.900, P= 0.004). The fibrosis level in primary myelofibrosis group was mainly "grade 2" to "grade 3", and the non primary myelofibrosis group was mainly "grade 1" to "grade 2", and the differences of myelofibrosis degree component between them were also statistically significant (χ2= 12.692, P= 0.003). Conclusions Hematological malignancies, especially primary myelofibrosis, are the common causes of bone marrow "dry pumping". Bone marrow histology, reticular fiber staining and selective immunohistochemical staining are of great significance in the etiological diagnosis.

ABSTRACT

Objective:To investigate the effects of Exendin-4 (Ex-4) on the expressions of lipid metabolism related genes in the human liver cancer HepG2 cells with insulin resistance (IR),and to elucidate the effect of Ex-4in improvement of IR.Methods:The HepG2 cells in logarithmic growth phase were induced into IR model with high concentration of insulin,then divided into control group (HepG2 cells),IR group (HepG2 cells were treated with insulin,HepG2-IR cells),and Ex-4 group (HepG2-IR cells were treated with Ex-4).Glucose oxidase (GOD-POD)kit was used to detect the consumption of glucose.The cell morphology and intracellular lipid drip formation were observed by Oil red O staining.The triglyceride (TG) level in cells was detected by kit;qRT-PCR was used to detect the mRNA expression levels of acetyl-CoA carboxylase (ACC),fatty acid synthase (FAS),sterol regulatory element-binding protein-1c (SREBP-1c) and apolipoprotein B100 (apoB100).Results:Compared with control group (HepG2 cells),the glucose consumption in the HepG2-IR cells in IR group was significantly decreased (P<0.01).Compared with IR group,the glucose consumption in the HepG2-IR cells in Ex-4 group was increased (P<0.05).The Oil O red staining results showed that compared with control group,the fat percentage in the HepG2-IR cells in IR group was increased (P<0.05);compared with IR group,the fat percentage in Ex-4 group was decreased (P<0.05).Compared with control group,the level of TG in the cells in IR group was significantly increased (P<0.01);compared with IR group,the level of TG in the cells in Ex-4 group was significantly decreased (P<0.05).The qT-PCR results showed that compared with control group,the expression levels of ACC FAS and SREBP-1cmRNA in the cells in IR group were increased (P<0.01),and the expression level of apoB100 mRNA was decreased (P<0.05);compared with IR group,the expression levels of ACC,FAS and SREBP-1c mRNA in the cells in Ex-4 group were decreased (P<0.05),and the expression level of apoB100 mRNA was increased (P<0.01).Conclusion:Ex-4 can regulate the expressions of lipid metabolism related genes in the HepG2 cells and improve IR.

ABSTRACT

Objective To investigate the clinical and laboratory features of IgG-2κ light chain multiple myeloma. Methods The clinical data and laboratory results of 2 multiple myeloma (MM) patients with IgG-2κ light chain were analyzed and the related literatures were reviewed. Results Two male and 1 female patients were 50-82 years old and mainly suffered with backache, infection, anemia and renal dysfunction. Multiple osteolytic bone destruction was detected in X-ray as well as magnetic resonance imaging (MRI). The level of serum IgG was normal, slight or obviously increased, but the levels of IgA and IgM were decreased. The levels of κ light chain in serum and urine were both increased significantly, and Bence-Jones protein was positive. Double M protein peaks of serum in γ area were detected by protein electrophoresis in 2 patients. A single band of IgG and double bands of light chain κ were revealed by immunofixation electrophoresis. Bone marrow smear showed that abnormal plasma cells were increased obviously. One patient gave up chemotherapy because of lung infection, acute left heart failure and acute renal failure, the others 2 patients achieved partial remission and stable disease by receiving DVD and VAD chemotherapy. Conclusions IgG-2κ light chain MM lacks typical clinical presentation, but some laboratory characteristics may be different from those of IgG-κ light chain. Further researches are needed to confirm whether or not it belongs to biclonal MM.

ABSTRACT

Objective To observe the changes of serum and fecal taurine-conjugated bile acid levels and its association with glucose metabolism during the spontaneous development of type 2 diabetes in OLETO rats.Methods Twenty male OLETF rats(4 weeks old)were included and 10 male LETO rats of the same age were used as the normal control group.OLETF rats were fed with high fat diet whereas LETO rats were fed with normal diet.Serum and fecal taurine-conjugated bile acid levels of OLETF rats were tested at different stage of diabetes including baseline, normal glucose tolerance, impaired glucose tolerance and diabetes periods, and the association of taurine-conjugated bile acid level with body weight, blood glucose, and glucose-regulating hormones were also investigated.Results Compared with LETO rats, the baseline serum levels of taurine-conjugated bile acid in OLETF rats did not change, but the levels of fecal taurine-conjugated bile acid including taurine-conjugated chenodeoxycholic acid(TCDCA), taurocholic acid(TCA)and taurine-conjugated deoxycholic acid(TDCA)were significantly decreased [(14.25±7.18 vs 0.90±0.31)mg/kg,(7.12±4.14 vs 1.30±0.35)mg/kg,(4.30±1.78 vs 1.02±0.14)mg/kg, all P<0.01].During the development of diabetes, the fecal levels of TCDCA, TCA and TDCA were still lower than those in the control rats.TDCA was negatively associated with the level of fasting blood glucose(r=-0.470, P=0.032),but positively associated with the serum level of glucagon-like peptide(GLP)-l(r=0.406, P=0.044).Conclusion The decrease of intestinal taurine-conjugated bile acid level is involved in the development of diabetes in OLETF rats.Intestinal TDCA may regulate the secretion of GLP-1 by paracrine pathway.

ABSTRACT

Objective To measure the volume of pericardial adipose tissue (PAT) by a photons dual-source computed tomography (CT) (Somatom Definition Flash CT), and to analyze and evaluate the effects of pericardial adipose tissue volume on coronary artery imaging. Methods There were 123 patients with suspected coronary heart disease (CHD) who underwent CT coronary angiography (CTA) from February 2016 to August 2016 in our hospital were enrolled. According to the body mass index (BMI) of the patient, the tube current was set in the range of 100~400 mA and tube voltage was 100~120 kV. All the patients were divided into six groups from low to high based on the measured pericardial adipose tissue volumes. The coronary CTA imaging quality was evaluated by subjective scoring and objective comparison by calculating the signal-to-noise ratio and comparing the contrast-to-noise ratio. Statistical analysis was performed using Kmskal-Wallis rank sum test and Pearson correlation analysis. Results There was no significant difference in subjective scoring between different PAT groups (all P>0.05). There was no significant correlation between PAT content and signal-noise ratio and contrast noise ratio (P>0.05). Conclusions The pericardial adipose tissue volume has no significant effect on coronary artery CTA imaging.

Objective To investigate the clinical characteristics of hodgkin's lymphoma (HL) patients . Method The clinical data of HL patients in our hospital from January 1, 2003 to December 31, 2012 were analyzed retrospectively. The contents of investigation included gender, age, pathological type, first symptom, the incidence locations, clinical stages, extranodal involvement locations, ECOG and treatments.Results A total of 143 cases of HL were included, the M/F ratio was 2.5:1 with a median age of 37 years (11~73years) . The most frequent subtypes of HL were mixed cellular (34.3%) and nodular sclerosis (28.0%) . The nodal lymphomas accounted for 97.2%, while the extranodal lymphomas occupied 2.1%. The nodal lymphomas with extranodal invasion accounted for 29.5%, and the common locations were spleen (14.4%), lung (5.8%) and bone (5.0%) . The most clinical stage of all patients was stage Ⅰ/Ⅱ (60.8%), The differences were no significant in gender, pathological type, first symptom, the incidence locations and the effective rate of the first-line treatment (P>0.05), while were statistically significant in B symptoms, ECOG and treatments betweenⅠ/Ⅱgroups andⅢ/Ⅳgroups (P<0.05) . Conclusions In this group,the incidence of HL patients were mainly found in middle-aged people, males were lager than females.The pathological type, first symptom and the incidence locations had no obvious effect on clinical stage. The clinical stage had no obvious influence on the effective rate of the first-line treatment.

ABSTRACT

[Summary] A total of 128 individuals with type 2 diabetes underwent continuous glucose monitoring for 3 consecutive days.The dawn phenomenon was defined by three different parameters according to the previous research:(1)the absolute increase of glucose level from nocturnal nadir to prebreakfast value(?G) above 20 mg/dl;(2)?G above 10 mg/dl;( 3 ) insulin requirement increased at least 20%.The participants were secondarily separated by presence/absence of a dawn phenomenon based on the definitions above.The impact on blood glucose fluctuation of different groups was assessed according to the standard deviation of blood glucose( SDBG) , the area under curve above 10 mmol/L ( AUC ) , and the mean amplitude of glycemic excursions ( MAGE ) , etc.The frequencies of dawn phenomenon were 64.8%(?G≥20mg/dl), 85.2%(?G≥10 mg/dl), and 59.4%(rise in insulin requirement≥20%)respectively.The impacts on SDBG, AUC, MAGE, and MODD were without statistical difference(P>0.05) between the presence and absence of the dawn phenomenon patients when?G≥10 mg/dl.However, the differences reached statistical significance(P<0.05) when ?G≥20 mg/dl and the increase in insulin requirement≥20%. Besides, the incidence of dawn phenomenon was positively correlated with HOMA-IR, HbA1C , and free C-peptide.Dawn phenomenon is a very frequent event in type 2 diabetes and not only impacts the overall glycemic control but also exaggerates glucose fluctuation.To be clinically relevant, ?G≥20mg/dl should be taken as the quantitative criterion of the dawn phenomenon.

ABSTRACT

Objective:To study the effects of dental fluorosis on the tooth movement following extraction of adjacent tooth in adoles-cents for orthodontic treatment.Methods:50 adolescents were divided into mild(n =1 3),moderat(n =1 0),severe(n =7)and healthy control(n =20)groups according to the fluorosis severity using Dean's index.4 4 were extracted and appliance was applied. X-ray image was taken 1 week and 1 ,2 and 3 months after appliace activation.Tooth movement was measured cephalometrically by Auto CAD.Results:Tooth moving distance was more(P <0.01 )and the alveolar bone resorption area was smaller in fluorosis group than those in healthy control group(P <0.01 )1 ,2 and 3 months after appliance activation.The moving distance was negativelly relat-ed with bone remodling area(P <0.05).Conclusion:Adolescent dental flurosis play an important role in bone remdodeling in extra-tion sites and tooth movement during orthodontic treatment.

ABSTRACT

Objective The purpose of this study was to study the correlation between dental fluorosis, saliva and plaque fluoride levels and urinary fluoride values in adolescents dental fluorosis. Methods A middle school was chosen as a survey point in the study. Two hundred adolescents were examined the degree of dental fluorosis by Dean's method. These adolescents were divided into four groups according to the severity of fluorosis (n = 52, 40, 28 and 80). Fluoride ion specific electrode was used to measure the fluoride levels in dental plaque, saliva, urinary and drinking water. The differences were analyzed b y ANOVA. Correlation of the fluoride levels between dental plaque, saliva, urine and the degree of dental fluorosis were analyzed by the method of multiple linear regression. Results The average fluoride content of drinking water was (2.20 ± 0.40) mg/L. Compared with controls, the fluoride concentrations in dental plaque, saliva and urine were higher in light, medium and severe dental fluorosis groups [(1.55 ± 0.88), (1.94 ± 0.77), (2.74 ± 0.83) than (0.32 ± 0.20) mg/L; (4.44 ± 1.62), (8.09 ± 0.93), (10.72 ± 0.99) than (0.02 ± 0.01) mg/L;(31.77 ± 6.09), (57.98 ± 1.83), (65.98 ± 2.78) than (13.06 ± 2.11) μg/g, all P<0.05]. Urinary fluoride was correlated with fluoride in saliva and dental plaque (r=0.245, 0.440, all P<0.05). Saliva fluoride was correlated with fluoride in dental plaque (r=0.849, P<0.01). The degree of dental fluorosis was correlated with fluoride in urine and saliva (r = 0.497, 0.896, 0.924, all P< 0.01). The multiple linear regression equation between fluoride in urine and the degree of dent al fluorosis, fluoride in dental plaque and saliva was as follow: y = 1.357 + 1.618x1 + 0.001x2 - 0.331x3 ± 0.69. Conclusions The metabolism of fluoride in body is related with oral fluoride repository in adolescents dental fluorosis. Fluoride in urine is influenced by plaque fluoride level, saliva fluoride concentration and the degree of dental fluorosis.

ABSTRACT

The recent explosive outbreak of Zika virus (ZIKV) infection has been reported in South and Central America and the Caribbean. Neonatal microcephaly associated with ZIKV infection has already caused a public health emergency of international concern. No specific vaccines or drugs are currently available to treat ZIKV infection. The ZIKV helicase, which plays a pivotal role in viral RNA replication, is an attractive target for therapy. We determined the crystal structures of ZIKV helicase-ATP-Mn(2+) and ZIKV helicase-RNA. This is the first structure of any flavivirus helicase bound to ATP. Comparisons with related flavivirus helicases have shown that although the critical P-loop in the active site has variable conformations among different species, it adopts an identical mode to recognize ATP/Mn(2+). The structure of ZIKV helicase-RNA has revealed that upon RNA binding, rotations of the motor domains can cause significant conformational changes. Strikingly, although ZIKV and dengue virus (DENV) apo-helicases share conserved residues for RNA binding, their different manners of motor domain rotations result in distinct individual modes for RNA recognition. It suggests that flavivirus helicases could have evolved a conserved engine to convert chemical energy from nucleoside triphosphate to mechanical energy for RNA unwinding, but different motor domain rotations result in variable RNA recognition modes to adapt to individual viral replication.

ABSTRACT

Malnutrition is one of the most important factors for the prognosis of patients with gastric cancer,but the mechanism of malnutrition is unclear at present. These patients are at risk of nutritional deple-tion from anorexia,dysphagia,and may be associated with the presence of two or more of the following cyto-kines with metabolic activity such as TNF-α,IL-1,IL-6,interferon,leptin,nuclear factor-κB and so on, which are secreted by the host-tumor interaction. The cytokines constitute a sophisticated regulatory network. Therefore,the study of the relationship between malnutrition and related factors in patients with gastric cancer may improve the nutritional status and prognosis of these patients.
